myelodysplastic syndromes (MDS) are a heterogeneous group of clonal hematological diseases. In addition to defects in hematology and stem cell progenitors, dysfunction of the bone marrow microenvironment (BMM) participate in the pathogenesis of MDS. In addition, deregulation of the immune response by pro-inflammatory response force in low-risk MDS, whereas predominates immunosuppression in high-risk MDS.
mesenchymal stroma (MSC), a part of the BMM, which is characterized by the growth of plastic adherent and multipotentiality. They exhibit immunomodulatory properties and maintain hematopoiesis. There is conflicting evidence about the status of MDS. The purpose of this study was to characterize the MDS-MSC and evaluate the effect of 5-Azacytidine.The MSC from MDS patients and controls were cultured and marked in accordance with the International Society of Cell Therapy recommendation.

We evaluated the effects of 5-azacytidine at MSC cytokine production.We including 35 MDS patients and 22 controls. MSC from patients and controls were cultured and characterized. MSC of patients showed morphological differences, but there are no differences in the immunophenotype or multipotentiality. Interleukin 6 (IL-6) is the major cytokine secreted MSC. MDS-MSC produce higher levels of IL-6, IL-17, interferon gamma, or interferon-γ (IFN-γ), and tumor necrosis factor alpha (TNF-α).
In vitro treatment of 5-azacytidine induced a significant decrease in the production of IL-6 by MDS MDS-MSC-MSC.The showed increased production of pro-inflammatory cytokines. In vitro treatment with 5-azacytidine causes a significant reduction in IL-6 production by MDS-MSC, restoring the levels of IL-6 to that found in control. MSC produced inflammatory cytokines involved in the pathogenesis of MDS, which represents future potential therapeutic target. In addition, the 5-azacytidine may have the effect of stroma, modulate immune responses in MDS.

bacterial flagellin, is recognized by the cell surface receptor pulses like (TLR) 5, is a potent activator of many types of cells, leading to activation of the innate or adaptive immunity, which is important in regulating the fibrotic process. However, the exact role of TLR5 signaling in liver fibrogenesis are still unclear, and this study aims to explain the underlying mechanism.
Flagellin injected into hepatotoxin- and cholestatic liver fibrosis induced murine model. Flagellin-induced activation of TLR5 significantly reduce the severity of liver fibrosis. Interestingly, the level of expression of interleukin-1 receptor antagonist (IL1RN) and interferons (IFNB) rose sharply in the treatment of fibrotic liver flagellin. Consistently, in vivo activation of TLR5 signaling increased sharply IFNB and IL1RN expression in the liver.
In particular, the injection of flagellin significantly worsen the severity of liver fibrosis in IFN-α / β receptor-1 (IFNAR1) knockout mice. Furthermore, expression of the heart of IL1RN in liver fibrosis of IFNAR1 KO mice was significantly lower than that of wild-type mice. In support of these findings, the production of flagellin-mediated IL1RN not enough to alleviate the severity of fibro-inflammatory response in the hearts of IFNAR1-deficient environment.
