In this study, we focus on the role of serum interleukin 6 (IL-6) concentration in predicting 5-year cardiovascular mortality in hemodialysis patients using the dialyzer reuse. We low-flux measured serum concentrations of IL-6 in 236 hemodialysis patients (138 males and 98 women) to predict the 5-year cardiovascular mortality.
We assess the basic demographics of all patients who had a mean age of 44 years and a median duration of 38.5 months hemodialysis. We divided all patients into two equal groups based on serum IL-6 concentration: G1 (n = 118) with serum IL-6 concentrations <6.78 pg / L and G2 (n = 118) with serum IL-6 concentration ≥ 6 , 78 pg /L.After 5 years of follow-up, 45 patients died of cardiovascular causes (19.1%).
lipid disorders, hemoglobin, serum albumin, β2-M, and IL-6 concentration is an independent risk factor for predicting cardiovascular mortality during the 60-month follow-up in patients on hemodialysis. Based on Kaplan-Meier analysis, we realized that patients with higher concentrations of interleukin 6 (G2) had a cardiovascular mortality rate was significantly higher than in G1 patients (log-rank test p <0.001).
Serum IL-6 concentration was a better predictor of 5-year cardiovascular mortality than the high-sensitivity C-reactive protein in hemodialysis patients using reuse the dialyzer low-flux (AUC = 0.818; p <0.001; cut-off value: 8.055 pg / mL, Se = 77.8%, Sp = 78.5%). Serum IL-6 concentration was a better predictor of cardiovascular mortality compared 5 years of high sensitivity C-reactive protein in maintenance hemodialysis patients using low-flux dialysis reuse.
Interleukin 6 is a better predictor of 5-year cardiovascular mortality than high-sensitivity C-reactive protein in hemodialysis patients using reused low-flux dialyzers.
Identification of new splice variants of mouse and human interleukin – 5 .
Interleukins and their receptors expression are often regulated by alternative splicing. alternative isoforms of IL-5 receptor α-chain is well studied; However, no data on functional alternative splice variant of IL-5 has been reported today. In this study, we describe a new splice variants of mouse and human IL-5. The new form is found during the analysis of PCR products amplified from mouse lymphoid tissues that are different primer pair is designed for full-length clone MIL-5 ORF.
A single short isoform MIL-5 detected along with the full-length canonical Cona-induced mRNA in lymphoid cells isolated from spleen, thymus, lymph nodes and blood. It was 30-40 nt shorter, and less than the classical form. Analysis of additional sequences such as MIL-5, revealing that he has no exon-2 (δ2). Using RT-PCR with primers specific splice we obtain additional evidence for the expression of form δ2.
To verify whether the MIL-5δ2 transcripts translated into proteins, the coding sequence in accordance with the full form and δ2 of MIL-5 cloned into the expression plasmid. After transfection into human 293T cell lines, we found that the short form of MIL-5 proteins are expressed in cells and is secreted into the supernatant, but at a reduced rate than that detected for the full MIL-5 isoform.
Fluorescence microscope examination revealed a partial MIL-5δ2 translocation into the cytoplasm, while MIL-5 stayed mostly in the endoplasmic reticulum. This may explain why the rate of protein expression δ2 reduced. Using a similar set of experimental approaches, we received evidence that human IL-5 mRNA has δ2 splice forms (HIL-5δ2) as well. It can firmly be detected by RT-PCR in PHA-activated mononuclear cells isolated from peripheral blood of healthy individuals or patients with asthma.

Overall, our results suggest that human and mouse IL-5 mRNA alternative splice isoforms, which lost exon-2, however, expressed at the protein level. However, a more comprehensive study would be required to evaluate the expression of IL-5δ2, regulatory, biological function and clinical significance.
